Viruses are small infectious agents that can replicate only inside the living cells of an organism. Viruses infect all forms of life, including animals, plants, fungi, bacteria and archaea. They are found in almost every ecosystem on Earth and are the most abundant type of biological entity, with millions of different types, although only about 6,000 viruses have been described in detail. Some viruses cause disease in humans, and others are responsible for economically important diseases of livestock and crops.

Virus particles (known as virions) consist of genetic material, which can be either DNA or RNA, wrapped in a protein coat called the capsid; some viruses also have an outer lipid envelope. The capsid can take simple helical or icosahedral forms, or more complex structures. The average virus is about 1/100 the size of the average bacterium, and most are too small to be seen directly with an optical microscope.

The origins of viruses are unclear: some may have evolved from plasmids, others from bacteria. Viruses are sometimes considered to be a life form, because they carry genetic material, reproduce and evolve through natural selection. However they lack key characteristics (such as cell structure) that are generally considered necessary to count as life. Because they possess some but not all such qualities, viruses have been described as "organisms at the edge of life".

The common cold is an upper respiratory tract disease that mainly affects the nose, and sometimes the throat, larynx and sinuses. Over 200 viruses can cause colds, most commonly rhinoviruses but also coronaviruses, influenza viruses, adenoviruses and others. Adults catch an average of 2–3 colds a year and children 6–8, making it the most common infectious human disease. The economic costs are huge, with colds responsible for 40% of time lost from work in the U.S. Colds are described in the Egyptian Ebers papyrus, the oldest surviving medical text, written before the 16th century BCE.

Symptoms include cough, sore throat, runny nose, nasal congestion, sneezing and sometimes muscle aches and headache; fever is common in young children. Symptoms typically resolve in 7–10 days, although some can last up to 3 weeks. The immune response to infection, rather than tissue destruction by the virus, causes most of the symptoms. Transmission occurs via airborne droplets and by contact with nasal secretions or contaminated objects. The viruses that cause colds can survive for prolonged periods in the environment (over 18 hours for rhinoviruses). Hand washing can help to prevent spread. No effective antiviral treatment or vaccine currently exists.

The common cold is the most frequent infectious disease. Despite the advice to "consult your physician" no antiviral treatment has been approved, and colds are only rarely associated with serious complications.

Interferons are a group of signalling proteins released by host cells in response to viruses and other pathogens. They belong to a large class of proteins known as cytokines: molecules used for communication between cells to trigger the protective defences of the immune system that help to eradicate pathogens. More than twenty distinct interferon genes and proteins have been identified in animals, including humans.

When a cell is infected by a virus, several virus products, including glycoproteins and viral RNA, stimulate the cell to produce and release interferons. This causes nearby cells to heighten their defences against viral infection, and so interferes with viral replication. Some viruses, including Japanese encephalitis virus, dengue type 2 virus, human cytomegalovirus and Kaposi's sarcoma-associated herpesvirus, have evolved ways to resist interferon's antiviral activity. Interferons also have various other functions in regulating the immune system. Interferons and other cytokines are responsible for some symptoms of infection, such as fever, muscle pain and "flu-like symptoms".

The 1918–20 influenza pandemic, the first of the two involving H1N1 influenza virus, was unusually deadly. It infected 500 million people across the entire globe, with a death toll of 50–100 million (3–5% of the world's population), making it one of the deadliest natural disasters of human history. It has also been implicated in the outbreak of encephalitis lethargica in the 1920s. Despite the nickname "Spanish flu", the pandemic's geographic origin is unknown.

Most influenza outbreaks disproportionately kill young, elderly or already weakened patients; in contrast this predominantly killed healthy young adults. Contemporary medical reports suggest that malnourishment, overcrowded medical facilities and poor hygiene promoted fatal bacterial pneumonia. Some research suggests that the virus might have killed through a cytokine storm, an overreaction of the body's immune system. This would mean the strong immune reactions of young adults resulted in a more severe disease than the weaker immune systems of children and older adults.

Rotavirus

Rotavirus is a genus of double-stranded RNA viruses in the family Reoviridae. There are nine species A–I; rotavirus A, the most common, causes over 90% of infections in humans. Rotavirus also infects animals, including livestock. The virus is transmitted by the faecal–oral route, with fewer than 100 virus particles being required for infection. Rotaviruses are stable in the environment and normal sanitary measures fail to protect against them. Effective rotavirus vaccines are the main prevention method.

The virus infects and damages the enterocytes lining the small intestine, causing gastroenteritis (sometimes referred to as "stomach flu," although the virus is not related to influenza). A viral toxin, NSP4, is responsible for some of the pathology. Rotavirus is the most common cause of diarrhoea among infants and young children. Almost every child worldwide has been infected with rotavirus at least once by the age of five. In 2013, 215,000 children under five died from rotavirus infection, mostly in developing countries, and almost two million more become severely ill. Immunity develops with each infection and adults are rarely affected.

Françoise Barré-Sinoussi in 2008

Françoise Barré-Sinoussi (born 30 July 1947) is a French virologist, known for being one of the researchers who discovered HIV.

Barré-Sinoussi researched retroviruses in Luc Montagnier's group at the Institut Pasteur in Paris. In 1982, she and her co-workers started to analyse samples from people with a new disease, then referred to as "gay-related immune deficiency". They found a novel retrovirus in lymph node tissue, which they called "lymphadenopathy-associated virus". Their results were published simultaneously with those of Robert Gallo's group in the USA, who had independently discovered the virus under the name "human T-lymphotropic virus type III". The virus, renamed "human immunodeficiency virus", was later shown to cause AIDS. Barré-Sinoussi continued to research HIV until her retirement in 2015, studying how the virus is transmitted from mother to child, the immune response to HIV, and how a small proportion of infected individuals, termed "long-term nonprogressors", can limit HIV replication without treatment. In 2008, she was awarded the Nobel Prize, with Montagnier, for the discovery of HIV.

Gardasil human papillomavirus vaccine

Several human papillomavirus (HPV) vaccines, including Cervarix and Gardasil, have been approved to protect against infections with particular types of HPV, associated with cervical and other cancers. All vaccines protect against the high-risk HPV types 16 and 18. Gardasil is a quadrivalent vaccine that additionally protects against low-risk HPV-6 and -11, which are associated with most cases of genital warts. A second-generation nine-valent Gardasil vaccine protects against five additional high-risk HPV types. It is estimated that the vaccines may prevent 70% of cervical cancer, 80% of anal cancer, 60% of vaginal cancer, 40% of vulvar cancer and possibly some oropharyngeal cancers. Protection lasts for at least 8–9 years. Some advocate giving Gardasil to men and boys with the primary aim of protecting their female sexual partners; others consider vaccinating only women and girls to be more cost effective. The licensed vaccines are subunit vaccines, containing only the L1 capsid protein of the virus, which self-assembles into virus-like particles. They are not effective in people already infected with HPV. Research is ongoing into therapeutic HPV vaccines including the viral oncoproteins, E6 and E7, but none has yet been licensed.
